Multi - Unit Ribozyme - Mediated Cleavage of bcr - abl mRNA in Myeloid
نویسنده
چکیده
Chronic myelogenous leukemia is characterized by the Philadelphia chromosome, which at the molecular level results from the fusion of the bcr gene on chromosome 22 and the ablgene on chromosome 9. The bcr-ablfusion gene encodes a novel tyrosine kinase with transforming activity. In this study, we have synthesized a multi-unit ribozyme that targets bcr-ab/ mRNA. In vitro ribozyme cleavage reactions show increased cleavage efficiency of this multi-unit ribozyme compared with single or double ribozymes. The multiunit ribozyme was then transfected into murine myeloblasts

متن کاملBcr-abl Silencing by Specific Small-Interference RNA Expression Vector as a Potential Treatment for Chronic Myeloid Leukemia
Background: RNA interference (RNAi) is the mechanism of gene silencing-mediated messenger RNA degradation by small interference RNA (siRNA), which becomes a powerful tool for in vivo research, especially in the areas of cancer. In this research, the potential use of an expression vector as a specific siRNA producing tool for silencing of Bcr-abl in K562 cell line has been investigated. Methods:...
متن کاملRibozyme-mediated inhibition of bcr-abl gene expression in a Philadelphia chromosome-positive cell line.
The bcr-abl fusion gene is the molecular counterpart of the Philadelphia chromosome (Ph1) and is directly involved in the pathogenesis of Ph1+ leukemia. Inhibition of bcr-abl gene expression may have profound effects on the cell biology of Ph1+ cells, as recent experiments with antisense oligonucleotides have shown. In this study we have designed and synthesized a unique ribozyme that is direct...
متن کاملSpecificity of ribozymes against the bcr-abl mRNAs in vitro.
Chronic myelogenous leukaemia (CML) is a disease characterized by the presence of the Philadelphia chromosome and the k r d l fusion gene, which result from a reciprocal translocation between chromosomes 9 and 22. Two forms of bcrabl mRNA exist, b3a2 and b2a2, depending upon whether the translocation includes bcr exon 3 or not. The p2 lP-*’ protein, a deregulated tyrosine kinase, is implicated ...
متن کاملComparison of the specificities and catalytic activities of hammerhead ribozymes and DNA enzymes with respect to the cleavage of BCR-ABL chimeric L6 (b2a2) mRNA.
With the eventual goal of developing a treatment for chronic myelogenous leukemia (CML), attempts have been made to design hammerhead ribozymes that can specifically cleave BCR-ABL fusion mRNA. In the case of L6 BCR-ABL fusion mRNA (b2a2 type; BCR exon 2 is fused to ABL exon 2), which has no effective cleavage sites for conventional hammerhead ribozymes near the BCR-ABL junction, it has proved ...
